Abnormal subcellular distribution of GLUT4 protein in obese and insulin-treated diabetic female dogs BJMBR
Vargas,A.M.; Barros,R.P.A.; Zampieri,R.A.; Okamoto,M.M.; de Carvalho Papa,P.; Machado,U.F..
The GLUT4 transporter plays a key role in insulin-induced glucose uptake, which is impaired in insulin resistance. The objective of the present study was to investigate the tissue content and the subcellular distribution of GLUT4 protein in 4- to 12-year-old control, obese and insulin-treated diabetic mongrel female dogs (4 animals per group). The parametrial white adipose tissue was sampled and processed to obtain both plasma membrane and microsome subcellular fractions for GLUT4 analysis by Western blotting. There was no significant difference in glycemia and insulinemia between control and obese animals. Diabetic dogs showed hyperglycemia (369.9 ± 89.9 mg/dl). Compared to control, the plasma membrane GLUT4, reported per g tissue, was reduced by 55% (P...

Acute effect of different antidepressants on glycemia in diabetic and non-diabetic rats BJMBR
Gomez,R.; Huber,J.; Tombini,G.; Barros,H.M.T..
Diabetic patients have a 20% higher risk of depression than the general population. Treatment with antidepressant drugs can directly interfere with blood glucose levels or may interact with hypoglycemic agents. The treatment of depression in diabetic patients must take into account variations of glycemic levels at different times and a comparison of the available antidepressant agents is important. In the present study we evaluated the interference of antidepressants with blood glucose levels of diabetic and non-diabetic rats. In a first experiment, male adult Wistar rats were fasted for 12 h. Imipramine (5 mg/kg), moclobemide (30 mg/kg), clonazepam (0.25 mg/kg), fluoxetine (20 mg/kg) sertraline (30 mg/kg) or vehicle was administered. After 30 min, fasting...

Diniz,S.F.; Amorim,F.P.L.G.; Cavalcante-Neto,F.F.; Bocca,A.L.; Batista,A.C.; Simm,G.E.P.M.; Silva,T.A..
A closed fracture was performed on the left tibia of 3-month-old Wistar rats weighing 250 to 350 g that were either healthy (N = 24) or made diabetic with alloxan (N = 24) to investigate the effect of alloxan-induced diabetes on the course of bone fracture healing. Histomorphometric analysis of the fracture site was performed at 7, 14, 25, and 35 days. After 7 days, diabetic rats had significantly less cartilage (P = 0.045) and greater fibrous connective (P = 0.006) tissue formation at the fracture site compared to controls. In contrast, marked callus formation was seen in diabetic rats with significant osteogenesis (P = 0.011, P = 0.010, P = 0.010, respectively, for 14, 25, and 35 days) and chondrogenesis (P = 0.028, P = 0.033, P = 0.019) compared to...

Amylin induces hypoglycemia in mice Anais da ABC (AABC)
Guerreiro,Luiz H.; Silva,Daniel DA; Sola-Penna,Mauro; Mizurini,Daniella M.; Lima,Luís M.T.R..
Amylin is a 37-aminoacid pancreatic protein that exerts control over several metabolic events such as glycemia and lacticemia. Amylin has long been shown to induce increases in arterial plasma glucose. We decided to investigate whether amylin plays additional roles in the glucose metabolism. We evaluated glucose homeostasis using whole blood from the tail tip of fasting, conscious, unrestrained normal and streptozotocyn-induced diabetic mice following subcutaneous administration of mouse amylin. Subcutaneous injection of 1 μg mouse amylin caused a transient decrease in whole blood glucose in both normal and diabetic mice in the absence of insulin. The blood glucose levels were lowest approximately 2 hours after amylin administration, after that they...

Biological response of spontaneously hypertensive rats to the streptozotocin administration BABT
Costa,Virginia Alice Vieira da; Vianna,Lucia Marques.
The sensitivity of adult spontaneously hypertensive rats (SHR) to the diabetogenic effect of streptozotocin (STZ) was studied. The animals were subdivided into three groups: control (citrate buffer), streptozotocin 40 mg/kg or 50 mg/kg, and general biologic parameters were analyzed, in addition to systolic blood pressure, blood glucose and insulin levels determinations. Both doses were able to induce hyperglycemia above 300 mg/dl; however, 50 mg/kg provoked a more pronounced physiological alterations in body weight, diuresis, water and food intake. There was no change on systolic blood pressure with either dose. Results suggested that SHRs did not need doses of streptozotocin above 40mg/kg in order to produce diabetes probably because this strain was much...

Chloramphenicol restores the polymorphonuclear accumulation in carrageenin-induced pleurisy in diabetic rats Braz. J. Vet. Res. Anim. Sci.
MORAES,Julieta Rodini Engrácia de; MORAES,Flávio Ruas de; ENGRÁCIA FILHO,Jair Rodini.
We investigated the effect of chloramphenicol succinate (30 mg/kg, every 12 h for 4 days, ip) on the reduced polymorphonuclear leukocytes (PMN) accumulation in carrageenin-induced pleurisy (150 mug) in diabetic (alloxan, 40 mg/kg, IV) rats (Wistar, males, 180-320 g, n = 12). Chloramphenicol produced an increase (p<0.05) on PMN count in pleural exudate of 36% in normal animals. In the animals non-treated with chloramphenicol the diabetes state reduced (p<0.05) the accumulation of PMN cells by 45%. In diabetic animals treated with chloramphenicol the number of PMN in pleural exudate was similar to the one observed in the control group. Total and differential counts of leukocytes in the peripheral blood were similar between groups before and 4 h after...

Chlorophyll treatment combined with photostimulation increases glycolysis and decreases oxidative stress in the liver of type 1 diabetic rats BJMBR
Wunderlich,A.L.M.; Azevedo,S.C.S.F.; Yamada,L.A.; Bataglini,C.; Previate,C.; Campanholi,K.S.S.; Pereira,P.C.S.; Caetano,W.; Kaplum,V.; Nakamura,C.V.; Nakanishi,A.B.S.; Comar,J.F.; Pedrosa,M.M.D.; Godoi,V.A.F..
Photodynamic therapy (PDT) promotes cell death, and it has been successfully employed as a treatment resource for neuropathic complications of diabetes mellitus (T1DM) and hepatocellular carcinoma. The liver is the major organ involved in the regulation of energy homeostasis, and in pathological conditions such as T1DM, changes in liver metabolic pathways result in hyperglycemia, which is associated with multiple organic dysfunctions. In this context, it has been suggested that chlorophyll-a and its derivatives have anti-diabetic actions, such as reducing hyperglycemia, hyperinsulinemia, and hypertriglyceridemia, but these effects have not yet been proven. Thus, the biological action of PDT with chlorophyll-a on hepatic parameters related to energy...
